Evaluation of the Physiological Activity on the Skin and Identification of the Active Ingredient of Leaf Extract from Sanhyang Sandolbae (Pyrus ussuriensis) as a New Varie

Abstract
Recently, a new variety of the Pyrus ussuriensis, called ¡®Sanhyang¡¯, was developed through varietal improvement. In this study, seven ethanolic extracts were prepared from the dried leaves of Sanyang and six wild P. ussuriensis (PU) species, and the physiological activities on skin cells were compared. The extraction yield was higher in the leaf extract of Sanhyang (CS-PU; 32.7 mg/100 mg) than those of the wild species (23.7¡­28.9 mg/100 mg). The anti-inflammatory activity measured using LPS-stimulated RAW 264.7 showed that CS-PU at a dose of 100 ¥ìg/mL inhibited IL-1¥â (38.8% inhibition), IL-6 (67.1% inhibition), and TNF-¥á (54.3% inhibition) production significantly. An analysis of wrinkle improvement and moisturizing enhancement using HDF and HaCaT cells showed that CS-PU at a dose of 100 ¥ìg/mL decreased the metalloproteinase-1 level (44.5% inhibition) and increased the hyaluronic acid level (85.3%). The anti-atopy activity measured using TNF-¥á and IFN-¥ã-stimulated HaCaT showed that the thymus and activation-regulated chemokine level was reduced by 59.7% in the CS-PU group treated with 100 ¥ìg/mL compared to that of the atopy-induced control. HPLC and mass spectrometry showed that CS-PU contained 11.9 mg/g of chlorogenic acid and 9.7 mg/g of cynaroside. In conclusion, CS-PU has higher extraction yield and its promoting effects on the physiological activities on the skin appear to be outstanding compared to those of the wild PU species. Therefore, CS-PU has potential use as a functional cosmetic ingredient.
